This study developed and validated a hydrophilic interaction liquid chromatography–tandem mass spectrometry (HILIC-LC-MS/MS) method for the simultaneous determination of N-acetyl neuraminic acid (NANA) and N-glycolyl neuraminic acid (NGNA) in edible bird’s nest and bird’s nest products. Samples were hydrolyzed with 0.1 M hydrochloric acid at 90℃ for 1 h to release bound sialic acids prior to analysis. Chromatographic separation was performed on an XBridge HILIC column (150 × 4.6 mm, 3.5 µm) using a mobile phase consisting of ammonium formate/formic acid and acetonitrile under gradient elution. The analytes were detected by tandem mass spectrometry in positive electrospray ionization (ESI+) mode using multiple reaction monitoring (MRM). The method showed good linearity over the concentration range of 25 – 5000 ng/mL, with a correlation coefficient (r) ≥ 0.999. The limits of detection (LOD) and quantification (LOQ) were 0.37 and 1.23 mg/kg, respectively. The precision and recovery of the method met the AOAC validation requirements. The method was applied to the simultaneous determination of NANA and NGNA in edible bird’s nest and bird’s nest product samples. The results showed that NANA was the predominant sialic acid component, whereas NGNA was detected only at low levels in some samples. The application results demonstrated that the developed method met the requirements for the simultaneous determination of NANA and NGNA in edible bird’s nest and bird’s nest products.
