Tình trạng trộn trái phép các dược chất như sibutramin, furosemid và phenolphtalein vào thực phẩm giảm cân đang là vấn đề đáng lo ngại, ảnh hưởng nghiêm trọng đến sức khỏe người tiêu dùng. Nghiên cứu này nhằm xây dựng và thẩm định một quy trình phân tích đồng thời ba chất trên trong thực phẩm giảm cân bằng phương pháp sắc ký lỏng hiệu năng cao kết hợp đầu dò quang phổ diode (HPLC-PDA). Quy trình được tối ưu hóa sử dụng cột InertSUSTAIN Phenyl (250 mm × 4,6 mm, 5 µm), pha động acetonitril – acid phosphoric 0,5% (45:55), tốc độ dòng 1 ml/phút, bước sóng phát hiện 225 nm. Mẫu được chiết bằng ethanol 96% và xử lý đơn giản. Phương pháp được thẩm định theo hướng dẫn AOAC với các tiêu chí: tính chọn lọc, tuyến tính, độ đúng, độ chính xác, giới hạn phát hiện (LOD), giới hạn định lượng (LOQ) và ảnh hưởng của độ pha loãng. Kết quả cho thấy phương pháp có độ tuyến tính cao (R² > 0,998), độ thu hồi đạt yêu cầu (95–103%), LOD và LOQ thấp (20,2 – 37,9 µg/g). Ứng dụng phương pháp để khảo sát 19 mẫu thực phẩm giảm cân trên thị trường, phát hiện 4 mẫu chứa sibutramin và 2 mẫu chứa phenolphtalein. Phương pháp có thể triển khai tại các phòng kiểm nghiệm để kiểm soát chất lượng thực phẩm giảm cân, góp phần bảo vệ sức khỏe cộng đồng.
