Tóm tắt

Niken-Sepharose là một trong những giá thể thông dụng nhất trong việc tinh sạch protein tái tổ hợp mang His-tag (His-tag protein) dựa vào liên kết ái lực giữa trình tự polyhistidine và ion Ni2+. Tuy nhiên, giá thành của Sepharose được cho là khá cao, đặc biệt là cho các nghiên cứu về protein ở Việt Nam. Trong nghiên cứu này, chúng tôi lần đầu tiên chỉ ra rằng melanin, được tách chiết từ túi mực là phế phẩm trong ngành thực phẩm, có thể được sử dụng làm giá thể tiềm năng để tinh sạch các His-tag protein. Melanin và melanin sau khi hấp phụ ion kim loại đã được thử nghiệm để làm chất nền cho quá trình tinh sạch His-tag protein. Kết quả cho thấy protein tái tổ hợp mang His-tag VP28 trong dịch chiết protein tổng số đều được bắt giữ bởi các giá thể melanin hấp phụ ion kim loại Ni2+, Fe3+ và Zn2+ cũng như là giá thể melanin dạng đơn. Các thử nghiệm giải phóng His-tag VP28 ra khỏi giá thể được thực hiện trên chất nền melanin và melanin hấp phụ Ni2+ (Ni-melanin). Kết quả cho thấy protein này được giải phóng ra tương đối chọn lọc khi sử dụng đệm đẩy imidazole 250 mM ủ qua đêm. So sánh một cách tương đối với Ni-Sepharose thì melanin và Ni-melanin cho hiệu quả tinh sạch lần lượt là 38% và 18%. Hiệu quả tinh sạch này là chưa cao, tuy nhiên các nghiên cứu tối ưu quy trình có thể sẽ giúp tăng hiệu suất và tăng độ tinh sạch của His-tag protein.
